Objective:

The Herbarium VIES collection has about 46,500 specimens, of which approximately 2,000 correspond to bryophytes and fungi. The entire collection is digitalized, but for these groups it is essential to scan temporary histological laminas, spores or tissues to correct identifications. So, the microscopic photos for them are so important as the macroscopic photos available in virtual herbaria. Thus, the aim of this project is to scan the bryophytes and fungi laminas from the VIES Herbarium collection and make the virtual herbarium available, so that they can be consulted online by the experts.

Plan:

For fungi, histological sections of the dehydrated basidiomes will be made freehand using steel blades and then mounted in solution KOH 3%, Phloxin and Melzer reagent; or small portions of the gleba will be removed with a needle to scan the basidiospores. For exoperidium, small parts will be removed and sectioned and mounted on laminas and glass slides to scan the hyphae. For ascospores and conidia the anatomical preparations will be made in distilled water and solutions of lugol 2% and KOH 10%. For other structures specific bibliographies will be used.
For bryophytes, the scan will be performed in a 10x lens for the general gametophyte structure and for specific parts relevant to specimen identification. In mosses, the 10x, 40x and 100x lens will be used for gametophyte and for sporophyte. For liverworts, the general structure of the gametophyte will be scanned (10x), as well as the structures (100x). For the talose species, the scanned laminas will contain the thallus structure (10x) and its cross section (100x). For anthoceros, the thallus structure will be scanned (10x) and its cross-section and spore (100x).
The images will be uploaded to the INCT-HVFF (CRIA servers located at RNP Internet Datacenter, Brasília, Brazil) and Reflora (servers located at Internet Datacenter at the Rio de Janeiro Botanical Garden, Rio de Janeiro, Brazil) virtual herbaria servers and available with free and open access at http://inct.splink.org.br/ and http://vies.jbrj.gov.br/.

Timetable:

To scan all fungi collection, according to the methods cited, the execution timetable provides for a period of 3 years, with an average of 500 samples per year, as it requires specific preparations for each group.
For a bryophyte collection, an estimated execution timetable for a 1-year for scanning the entire collection.
Thus, in the first year of the project execution, 100% of the bryophyte collection will be scanned and 1/3 of the fungi collection, in the second year 2/3 of the fungi collection, and in the third year, 100% of the fungi collection.
